The Surface glycoprotein (S) gene is a protein encoding gene at the GU280_gp02 locus tag found in the severe acute respiratory syndrome organism coronavirus 2 (SARS-CoV2) virus. Wuhan-Hu-1 isolate, nat-host:Homo sapiens. The SARS-CoV-2 virus is a positive enveloped single-stranded RNA virus that causes coronavirus disease 2019 (COVID-19), the structural proteins of SARS-CoV-2 including protein S are important targets for vaccine development, antibody therapy and antigen-based diagnostic tests. This study aimed to analyze genetic variation in the surface glycoprotein (S) gene sequence in SARS-CoV-2 NCBI PopSet: 1843471817 using in-silico RFLP. The RNA sequences were cut using 2 types of restriction enzymes, namely BamH1 and BglII. The results of in-silico RFLP showed that there were genetic variations on the restriction site of the BamHI enzyme (Alleles A1 and A2 with allele frequencies of 0.125 and 0.875) and BgIII (Alleles B1 and B2 with allele frequencies of 0.875 and 0.125).
